Content not loading

Hi, I just install my files to the domain. I am able to access the site but there is a problem with an image file just beneath the slider, not loading.I have checked using the File Manager and confirm that the file is rightly placed with permissions set to 777.Can someone kind to help a newbie. - All images should have permissions of 644.
- Have you tried reuploading the images?
- If so, how did you upload them?

Thanks for the reply. The permission is set to 644 but still the images are not loading. I have uploaded using the file manager and to the public_html/mystuff/images folder. Using Firebug to point to the content, I can see the error message,
"Failed to load given URL" Anyway the images are in jpg format.

I am still unable to solve the problem. I have even reupload files to create a new website and the same problem is unresolved. The new address is http://vanni.x10.mx/vanni/
