How to create a Minecraft Server in a Subdomain? (example: play.site.com:0000)

You would not be able to host any kind of server on a free hosting account (not on any shared hosting for that matter), you'd need to have a VPS at the least for that.
If you have a server space where it is installed you can simply create a subdomain and point it to that server.

You cannot assign port numbers in a DNS entry.

What you can do is get some separate MC server hosting (https://www.google.com/search?q=minecraft+server+hosting&ie=UTF-8#q=minecraft+server+hosting) . Then get the IP of your MC host server. Then, go to your x10 site cpanel, hit "Advanced DNS zone editor", and add a new record:
Name: play.yourdomain.com
Type: A
Value: your MC host IP

Hit save, wait up to 48 hours, and bam, you have a minecraft server at play.yourdomain.com
